Kininogen Knockout Mice
This Kininogen (KNG1) knockout mouse model was created by targeted disruption of KNG1 using programed nuclease. Mutations are confirmed by PCR genotyping and Sanger sequencing. The complete absence of kininogen protein in the plasma of homozygous mice is confirmed by Western Blot. High molecular weight kininogen (HMWK) is a cofactor for activation of prekallikrein, Factor XII, and Factor XI. This mouse has an extended clotting time because of this deficiency in a critical component of the intrinsic pathway of the clotting cascade. Kininogen is the precursor of the vasodilator peptide bradykinin and genetic deficiency may also impact blood pressure, inflammation, and pain responses. This KNG1 KO mouse model will be useful for cardiovascular studies including intrinsic/contact pathway coagulation and hypertension research. These kininogen deficient mice are in breeding now and homozygous mice will be available soon.
